Look for Squash Vine Borers. Additional reading: hgic.clemson.edu You can slit the infested vine lengthwise (near the sign of the borer) and remove borers or kill them with a long pin or needle. You may have squash bugs. Look them up on the Internet. Supposedly they are difficult to kill, but I put garlic cloves, hot chile pepper and murphy's oil soap together and sprayed my plants. The nymphs dried died right away. I had to pick the adults off by hand and drop them into soap water. Cut off your lower leaves and stake your plants to make it difficult for the bugs to hide. ...more
