Wow, there's a tough question... I suppose you'd have to first decide whether to treat all similar operating systems (eg, Linux variants) distinctly or separately. Also, in the early days, each model of computer had its own OS written for it.
OS/360 was the first OS for a standard instruction set, evolving eventually into z/OS. Since those days, I count around 23 distinct classes of OS from various manufacturers and with widely varying capabilities.
However there are at least 177 distinctly named OSes, including the more obscure ones like Newton OS and DR-DOS.
