    2M is a larger glycoprotein present in high concentration in the human plasma (2 to 3.5 gm/liter). Major resources of the –2M are the liver cells, macrophages and the fibroblasts. The liver cells and the macrophages are also responsible for the clearance of the ---2M. The macrophages and the fibroblasts production explains in presence of ---2M in the tissues and in tumors. Two forms have been describe native and inactivated –2M. The major functions of ---2M are its nonspecific, protease-binding capacity and its more recently recognized cytokines binding activity.

    Although the antiproteases bind and wrap themselves around the enzymes, the later remain active and the –2M functions more like the modulating and transport vehicle. This can be compare to soldier inside a tank that can still manoeuvres the tank and fire the weapons but can not get the out until the hatch is open and may be destroyed at a given time in the process. The enzymes in this activated complex state cannot be easily demonstrated by ordinary techniques, but their action is demonstrable. It appears that documentation of does related enzyme action therefore indirectly confirms their presence in the serum.
