Are you paying for the bus's gas?  It seems that MTA should be paying for the gas and upkeep of the bus.  75,000 is a lot per year, and the benefits of it being a M-F day job makes it even better.

If you want to be a medical assistant, though, then you should pursue your dream, especially if it's something that you want to do and think you would enjoy.

If your concern is money, though, being a medical assistant isn't what you should do.  According to this  a medical assistant made about 28,000/year in 2004.  In that case, you should go to school for nursing, and then apply for high stress jobs in a hospital (instead of at a clinic--hospital nurses make more than clinical nurses).  I don't know how much nursing jobs pay, so even then you may be taking a pay cut, and you would be working weekends/holidays, etc.

It depends on you. But if your job help you alot maybe you shouldn't. But the decision is yours.

If you quit your job do it because you are following your dreams of becoming a medical assistant ,rather than because of the price of gas since you seem to enjoy your job as a bus driver.
