If you live in the UK you can travel to major destinations in South America by air from London-Heathrow Airport, such as to Sao Paulo's Guarulhos International Airport , Rio De Janeiro's Galeão International Airport and Buenos Aires (Ministro Pistarini International Airport). These flights are generally operated by British Airways.
Major airports in the US, however, offer the best points of departure for South America. For example, from Washington-Dulles International Airport, you can fly to Sao Paulo, Santa Cruz in Bolivia, Rio De Janeiro, Montevideo in Uruguay, Guatemala City, Caracas (Venezuela) and Buenos Aires. Dulles is a good gateway to South America, but you can certainly also fly from New York's JFK Airport, or from Los Angeles, where you will also find regular flights to Lima.

The easiest way is by airplane, I go to Quito,Ecuador every summer. So, I go from Chicago to Miami, and Miami to Quito.
