Read over the job description and any other details which are relevent to the post.

Ask yourself have you got the relevent experience or similar experience for the job. Do you have the qualifications or similar. If you lack qualifications sometimes employers will consider you anyway.
Tell the prospective employer why you fit the criteria for the post and if you don't why they should interview you regardless.

Write about relevent things in a well constructed way making sure you have the job specification to hand. Make sure your grammar and spelling are correct, if your not sure ask someone to go over the application with you.

Can you provide referees who will back your claim that your the right person for the job. Don't appear over confident this is off putting.
