The letter was telling Romeo of Friar Lawrence's plan to have Juliet fake her death so that they can run away together without anyone chasing them. But because Romeo never received the letter he didn't know that Juliet was only sleeping and not really dead. Which is why he drank the poison he bought on his way to her tomb. If he had only gotten the letter than Juliet, Romeo and Paris would have lived through the play.
