I am unsure when to lock in a mortgage interest rate for a new home and have been quoted 5.375 percent.

It is unlikely mortgage rates will go up in the next week. If you have to pay a fee for the lock, then why do it. If you don't have to pay a fee and are happy with the rate, lock it in.
