The PCI-E slot is a part of the motherboard, so if you don't have this slot then you will need to buy a new motherboard, and maybe new CPU and memory too.
This will involve a fair amount of work, and you should feel confident with electronics before attempting it. My advice would be to return the graphics card or try to exchange it for an AGP version (if that's what your motherboard supports - most likely, but check your manual first!)
