    The correct punctuation is:

    Ladies' Room
    Men's Room

    Ladies is the plural form of lady, the apostrophe indicates possession.
    Men is the plural form of man, the apostrophe indicates possession.

    Huh? Ladies and Men are not equivalent terms. The equivalent term for Ladies is Gentlemen and the equivalent term for Women is Men. The signs should either read Women's Room and Men's Room or Ladies' Room and Gentlemen's Room.
