The earliest and most reliable way to diagnose pregnancy is with ultrasound.  Pregnancy can be diagnosed as early as day 20-22 but more reliable at day 26.  Uterine enlargement can be palpated around day 26-42 as well depending upon number of puppies.  Fetal skeletons calcify at day 45 and can be noted on x-ray.
Abdominal enlargement often starts around week 4-5.  This is often when your dog's appetite should increase.  Nipple development begins toward the end of pregnancy and they may have a discharge 10-14 days before birth.
It is important to diagnose pregnancy early so you can provide good pre-natal care.
