The Jello won't set because of the enzyme bromelain in the fresh pineapple. Gelatin is made from animal products and the bromelain destroys the protein in the gelatin, preventing the Jello from setting properly.  You can use canned pineapple instead, which has been heated in the canning process (which destroys the bromelain).
